Two police officers must go undercover as college students to track down a dangerous drug supplier.
The story
Two years after their success in the 21 Jump Street program, officers Schmidt and Jenko are investigating narcotics trafficking. After failing to stop a group of dealers led by a mysterious kingpin known as 'The Ghost', they are assigned to a new undercover mission. They must infiltrate a college as students to locate the supplier of a deadly synthetic drug called 'WHY-PHY'. While navigating campus life, the officers must balance their investigation with the complexities of student life, including forming alliances with local students and navigating the social dynamics of the university environment to find the source of the drug.

Following their success in the 21 Jump Street program, officers Schmidt and Jenko are tasked by Deputy Chief Hardy to work for Captain Dickson at 22 Jump Street. They go undercover as college students to find the supplier of 'WHY-PHY', a drug that killed a student. Jenko befriends jocks Zook and Rooster, while Schmidt pursues an art student named Maya, who turns out to be the daughter of Captain Dickson. After identifying a potential dealer and facing internal tensions regarding their future as officers, they eventually track down 'The Ghost'. During a spring break confrontation at a beach, they ambush a meeting where the Ghost's daughter, Mercedes, is coordinating deals. After a chaotic chase involving a helicopter, the Ghost is killed by a grenade. The film ends with the officers reconciling and being assigned a new undercover mission at a medical school, followed by a series of fictional sequels shown in the credits.
