Popular rock and roll superstar Conrad Birdie receives an Army draft notice, devastating his teenage fans nationwide. Unsuccessful songwriter Albert Peterson schemes with his secretary and girlfriend Rosie DeLeon to have Conrad perform a song Albert wrote, "One Last Kiss," on The Ed Sullivan Show, followed by a farewell kiss with a randomly chosen high-school girl. This plan is intended to free Albert to marry Rosie, who is often hindered by his meddlesome mother, Mae. The town of Sweet Apple, Ohio, is chosen for the farewell performance. The lucky girl chosen is Kim MacAfee, whose high-school sweetheart Hugo Peabody is jealous. During the event, Conrad's popularity causes local women to faint, and Kim's father initially refuses the kiss. Albert eventually convinces him by revealing his secret as a biochemist with a miracle farming supplement. During rehearsals, Conrad kisses Kim, causing her to dump Hugo. However, a Russian ballet troupe's performance threatens to cut Conrad's time. Rosie intervenes by drugging the conductor's milk with one of Albert's pills to speed up the ballet. Just as Conrad is about to kiss Kim, Hugo runs onto the stage and punches him. Kim and Hugo reunite, and Albert is finally free to marry Rosie, while Mae reveals her own marriage to a widower.
